The primary releasing hormone of reproduction is gonadotropin-releasing hormone (GnRH). GnRH controls the release of follicle-stimulating hormone (FSH)
and luteinizing hormone (LH) from the pituitary gland, located at the base of the brain. These pituitary hormones regulate the production of estrogen and progesterone from the ovary. FSH stimulates the follicle’s growth, development, and function, while LH causes\= the follicle to rupture and the corpus luteum to develop. The female hormone estrogen is produced by the Graafian follicle on the ovary. Another hormone originating from the

A relatively new discovery in reproductive physiology is that follicular growth is not continuous in the bovine, but occurs in waves (2 to 4 waves per cycle). Each wave begins when the dominant follicle in the previous wave gains maximal size, at which time numerous small follicles begin a period of rapid growth. From this group of follicles, one follicle is allowed to grow to a much larger size than the others. This large follicle is called the dominant follicle, because it has the ability to regulate and restrict the growth of the smaller follicles. A few days after reaching maximum size, the dominant follicle begins to degenerate and die.

Bleeding disorders can be either congenital, present from birth, or acquired, developing later in life. Defects in blood clotting proteins typically result in delayed bleeding and deep tissue bruising, whereas platelet defects often lead to superficial bruises, nosebleeds, black stools, and prolonged bleeding at injection sites or during surgery. The kidneys remove nitrogenous compounds like urea, uric acid, and creatinine, as well as metabolic waste products, toxins, excess hormones, water, and electrolytes. Urine formation involves three main stages: glomerular filtration, tubular reabsorption, and secretion. The kidneys produce renal filtrate, which contains blood components that can be filtered out, and the body recovers 99% of the water and electrolytes from the filtrate. The renal corpuscle, comprising the Bowman’s capsule and glomerular capillaries, is responsible for filtration of blood plasma and producing renal filtrate. Its structural elements allow
for efficient filtration, with glomerular capillaries having a 100-fold greater permeability to water, small ions, and solutes. The glomerular capillaries also have increased pressure due to the smaller diameter of the efferent arteriole. The filtration membrane consists of endothelium, basement membrane, and podocytes. The proximal convoluted tubule in the kidneys plays a crucial role in reabsorption of water and electrolytes, including sodium,
chloride, potassium, calcium, and bicarbonate. It reabsorbs approximately 80% of filtered bicarbonate, with further reabsorption from the loop of Henle and distal convoluted tubule. This reabsorption regulates the body’s acid-base balance, contributing to the acidic nature of urine. The proximal convoluted tubule is the only region able to reabsorb glucose. The loop of Henle is a crucial region in the kidney, responsible for concentrating urine and facilitating water and electrolyte reabsorption. It regulates fluid volume, blood pressure, and acid-base balance through bicarbonate reabsorption. The loop consists of two main regions: the descending limb and the ascending limb.

Creatinine Clinical significance Creatinine is produced at a steady rate due to muscle catabolism and is not reabsorbed by the kidney tubules after filtration. Its measurement provides an indirect assessment of the glomerular filtration rate (GFR). Unlike urea it is not markedly affected by diet or any aspect of liver function and therefore the test is more specific for renal dysfunction

Sodium Clinical Significance Compared to potassium, sodium is distributed differently throughout the body. Due to the sodium pump mechanism, most sodium is extracellular. Compared to this, just 2% of potassium is extracellular; the remainder is intracellular. Astudy was conducted to establish the regeneration capacity of pseudobulb segments of a Dendrobium hybrid orchid (D. nobile L. x D. chrysotoxum L.) in vitro on full strength (MS) and half strength (½ MS) Murashige and Skoog medium. Pseudobulb segments having 0.3-0.5 cm length cut from the in vitro maintained hybrid orchid were inoculated on both media supplemented with different concentrations and combinations of NAA, BAP and Kn. On MS medium the pseudobulb segments failed to respond, however, in ½ MS medium the explants responded well. Responding explants were maximum (35%) on the medium ½MS + 0.1 mgl^-1 NAA + 0.5 mgl^-1 BAP. An average of two (2) shoot buds per explant were regenerated on ½ MS + 2.0 mgl^-1Kn and ½ MS + 0.1 mgl^-1 NAA + 1.0 mgl^-1 BAP. The regenerated plantlets grew healthily on subculture after separation from the pseudobulb segments.

Clonal crops belong to at least 34 families including herbs, shrubs, trees and vines. Clonal crop improvement dates back to 10,000 years ago. There has been conscious effort to discover and apply biological pesticides for the control of insect predation. Insecticidal proteins/ genes from Bacillus thuringiensis have been deployed as sprays and through transgenic route for crop protection against insect attack and have been successful in protecting the crop damage and environment pollution. However, impending fear of development of resistance to Bt proteins by insects has prompted search for new insecticidal proteins. In this context we are exploring and analysing components of insect immunity as targets for restricting insect feeding behaviour. Insects encounter several microbes during different stages of growth and development. Incidental entry of these microbes elicits a host defense response, which mainly consists of a phenoloxidase (PO) pathway. The invasion of the microbe is recognized by a specific protein, which activates the PO pathway consisting of prophenoloxidase (PPO) & prophenoloxidase activating enzyme (PPAE). Understanding the mechanism of insect immunity will allow us to discover potential targets for inactivating the mechanism of immune action in insects and make them vulnerable to natural microbial infection.

An agronomically important polyphagous pest, Heliothis armigera (Bollworm) was chosen in the present work for studying the components of insect immunity. H. armigera is the most important insect pest of the developing world accounting for heavy damage on various crops including cotton, vegetables, tobacco, potato, groundnut, chickpea and pigeon pea. We have isolated full-length cDNA encoding prophenoloxidase (PPO) and a partial cDNA encoding PPAE from haemocytes of 5th instar larvae of H. armigera. The open reading frame (ORF) of PPO gene is 2.1 kb, which encodes for a 698 amino acid protein with a molecular mass of 77 kDa. Computer-assisted analysis revealed the presence of 60 amino acid long propeptide at the N terminus of protein and 2 copper binding motifs but no signal peptide was detected. The gene was cloned and expressed in a bacterial expression vector pET43b under the induction of IPTG. The SDS-PAGE analysis showed that the protein is expressing in the supernatant in its native form. The expressed enzyme is catalytically active on 4-Methylcatechol upon activation by cetyl pyridinium chloride (CPC). The regulatory role played enzyme in insect immunity is being examined.

ABSTRACT

Production of cardamom in India has been greatly affected by Cardamom mosaic virus (CdMV). The conventional breeding program to develop resistance is a lengthy process. An alternative approach is to develop transgenic cardamom through introduction and expression of viral sequences in the plant. The coat protein (CP) gene of all the CdMV isolates have been cloned in pGEMT vector and sequenced. Sequence comparison studies of the eight isolates of CdMV in their CP and 3’UTR showed that, Kursupara isolate is highly variable from the rest of the isolates (Jacob et al., 2003). Hence the present study was undertaken to introduce the Kursupara CP gene of CdMV into cardamom plants in order to develop pathogen-derived resistance in cardamom. The CP gene of the Kursupara isolate of the virus was amplified with and without the 3’untranslated region (3’UTR) from the viral clone. These PCR products were cloned into the pXcmkn12 vector and sub cloned in to a plant expression vector (pAHC17) at the BamHI site, under maize ubiquitin promoter. Work is in progress towards the biolistic transformation of cardamom with these constructs.

The cry1A genes of Bacillus thuringiensis (Bt) are widely used to engineer lepidopteran insect resistance in crop plants including horticultural crops. Continuous exposure of a single kind of Bt toxin can lead to resistance development in insect pest. Due to the difference in structure and unique insecticidal mechanism, cry2A genes are promising candidates for management of resistance development. Since India is rich in biodiversity and genetic resources, indigenous Bt strains are valuable tool for identification of novel cry genes. The present study describes the cloning of cry2Ab from a new indigenous strain of Bt, 14RI. The presence of cry2Ab gene in Bt strain 14RI was confirmed by PCR with primers specific to its internal region. Primers corresponding to the flaking region of cry2Ab were used for amplification of cry2Ab orf from the Bt strain 14RI. The amplified cry2Ab orf was first cloned into the T/A cloning vector, and then fused to the downstream of the cry2Aa promoter along with its orf1 and orf2 in the Bt shuttle vector pHT1301. Acrystalliferous Bt strain 4Q7 has been transformed with recombinant pHT1301 to express the cry2Ab of Bt strain 14RI.

ABSTRACT

The present study was conducted to screen fungal isolates for chitinolytic activity and to clone full-length endochitinase gene from fungus. Among the ten isolates tested (Trichoderma harzianum 1, 2, 3, Trichoderma koningii, Trichoderma virens, Trichoderma viride, Aspergillus oryzae Metarshizium verrucasia, Bauveria bassiana, Nomuraea releyi), Trichoderma koningii showed higest colloidal chitin hydrolysis when screened in Mandels and Reese (1969) salts supplemented with colloidal chitin as the sole carbon source followed by Aspergillus oryzae. Nomuraea releyi showed least chitinolytic activity. Other isolates were on par with each other. The use of diagnostic primers showed specific amplification of 260 bp in Trichoderma harzianum isolates 1,2 and3, Trichoderma koningii and Trichoderma virens, which confirmed presence of endochitinase gene.

Attempts were made to clone full-length endochitinase gene from isolates showing chitinolytic activity. A unique amplicon of 1.7Kb from Trichoderma virens was cloned into linear pTZ57R/T vector with T over hangs. The confirmation of clones was made through blue/white colony assay, plasmid mini-preparation and PCR amplification of these plasmids, sequencing and BLAST search at NCBI. Among the clones confirmed, pSAV804 was sequenced that showed 98% homology with other reported endochitinase genes from Trichoderma spp at DNA and protein level. 85.7% of the protein is conserved. The ORF of the sequence comprises of 822 bases, which deduces 274 amino acids. The position of primer sequence in the insert and BLAST results confirmed the reverse orientation in the vector. The endochitinase gene now needs to clone into the expression and plant expression vectors for further studies.

Introduction

Only after the discovery of restriction endonucleases it became possible to digest any DNA molecule at a specific site and any two pieces of DNA could be joined together with the help of another modifying enzyme, the DNA ligase. Consequently, a new branch of molecular biology, namely genetic engineering has been developed. With the help of genetic engineering techniques, it is now possible to identify and isolate the gene (for a protein of interest and insert it into the genome of another organism with the help of a suitable vehicle. The foreign gene thus cloned, can be amplified to produce millions of its copies. The insert gene can be expressed by the host to synthesize the gene product. The technique can also be used for understanding the regulation of expression of the cloned gene. The possibilities are endless and probably sky is the only limit. In this chapter we will briefly discuss the basic principles and protocol involved in gene cloning.

Clostridia infections are not common in birds and are not a significant source of infection for man and animals.

The genus Clostridium belongs to the family Clostridiaceae. The genus name was originated from the Greek word ‘kloster’ means spindle, referring to their individual rod shape. Clostridia are gram positive, large, sporulating anaerobic rods (except C. piliforme which is gram negative). At present, there are more than 200 numbers of species under the genus Clostridium. Among them, 15 species can produce potent toxins and are considered as pathogenic for animals and human. The animal pathogenic Clostridia can be classified as neurotoxic, histotoxic and enteric.

The genus Clostridium consists of gram positive, spore forming anaerobic bacilli. The spores are wider than the bacillary bodies, giving the bacillus a swollen appearance, resembling a spindle. Hence, the name clostridium (Kloster- meaning spindle) was given.

Clouds form when air becomes cooler until the water vapour present in it condenses to liquid form. The condensation over oceans occurs around solid particles like salt, dust, smoke etc. suspended in the atmosphere. The type of clouds depends upon temperature and wind. The clouds are classified based on physical appearance and the height at which clouds are formed (Fig. 7.1).

Cloud computing and Edge AI provide complementary infrastructure for intelligent livestock management. Cloud platforms offer scalable storage, centralized analytics, model training and integration of data from sensors, cameras, automated feeders and farm-information systems. Edge AI processes information close to the farm, reducing latency, bandwidth consumption and response time. This chapter explains their architectures, workflows and applications in dairy, poultry, swine and small-ruminant production. Uses include disease detection, estrus prediction, behavioural monitoring, feeding and environmental control. Hybrid cloud-edge systems balance centralized intelligence with local autonomy. Abstract

Cloud computing is becoming a technology mature enough for its use in genome research experiments. The use of large datasets, its highly demanding algorithms and the need for sudden computational resources, make large-scale sequencing experiments an attractive test-case for cloud computing. So far I have seen cloud computing demonstrated using R. However, it remains to be seen a rigorous comparison of its performance using a BLAST search and its ability to cope with ever-increasing databases and open source frameworks such as bioperl or bioconductor.

Clouds are the most important form of suspended water droplets caused by condensation. The World Meteorological Organisation defined cloud as a visible aggregate of minute particles of water or ice or both, in the free air. This aggregate may include larger particles of water or ice and particles such as those present in fumes, smoke or dust. The typical size of these water droplets and ice crystals range from less than 1 µm to 50 µm. When surrounded by billions of other droplets or crystals, they become visible as clouds. In the atmosphere sufficient moisture is required to allow condensation to take place. Then, moisture needs a suitable surface upon which it can condense. In free air, condensation begins on hygroscopic nuclei such as aerosols. These are microscopic particles and include dust, smoke, salts and chemical compounds. Nuclei range in size from 0.001 µm radius to over 10 µm. On an average, oceanic air contains one million condensation nuclei per litre and land air holds some 5 or 6 million. On hygroscopic particles such as sea salts, condensation can begin before the air is saturated. Supersaturation in clouds rarely exceeds one per cent. On average, clouds contain only four per cent of the total water in the atmosphere at any one time. Almost all clouds exist in the troposphere, although the tops of some severe thunderstorms occasionally extend to the stratosphere. Clouds move along with the wind and remain stationary in calm winds. They grow vertically and horizontally, depending on the temperature of the cloud and its ambient temperature. They may dissipate or precipitate. Under favourable atmospheric conditions, they burst and fall as rain. Clouds complete the following functions.

5.1 Clouds
Clouds are visible masses of tiny water droplets or ice crystals suspended in the Earth’s atmosphere. These droplets or crystals are so small and light that

Cloud usually consists of very small droplets of water which cannot fall appreciably below the cloud base without evaporation. In this condition clouds are stable. Rain drops are about a million times heavier than cloud drops and rain develops only if the cloud droplets grow by some mechanism. Cloud Seeding mechanism depends on the difference in vapour pressure over ice and super cooled cloud, water evaporates of the droplets and migrate to the ice crystal, which grows and begins to fall increasing their growth rate by coalescence with other droplets and eventually melting and falling as rain drops. Presence of ice nuclei determines whether ice crystals form in a supercoold cloud or not. Ice crystals form on naturally occurring nuclei at temperatures of – 10⁰ to 30⁰ C. At warmer temperatures, either the efficient ice-forming nuclei are often deficient in the atmosphere or the cloud may occur with tops supercooled but not to an extent which activates the ice nuclei available. It should be possible to initiate precipitation by introducing ice crystals or by supplying artificial nuclei.
