Bael (Angle marmelos) is an important arid fruits belongs to family Rutaceae and consists 2-3 species of which only one is in cultivation. It is important indigenous fruits of India, drought hardy tree, flowering and fruiting coincides with the onset of monsoon and fruits mature before commencement of summer. The tree shed off entire leaves during hot summer and fruits are exposed to direct sunlight, which favors ripening of fruits.

Demonstrations
Demonstration means showing by doing. The basic principle of Demonstration is learning by seeing and doing. The concept of demonstration was given by Dr. Seeman A. Knapp. Farmers like to see how a new idea works, both purposes can be achieved by means of a farm demonstration. A good, practical demonstration is an invaluable method in extension work. The demonstration is a particularly powerful method to use with farmers who do not read easily. There are two principal types of demonstration used by extension agents - method demonstration and result demonstration.

Radio script should be prepared in such a way that listener should feel that somebody is talking with him in a friendly manner and with informal style. For this purpose:
1. Collect the material from your experience.
2. Keep the purpose of the script and audience in mind.
3. Decide the method of presentation of information.
4. Considering the time available for the talk, the script should be prepared.
5. Generally we talk two words per seconds and 8 to 10 lines are required per minute having 5-6 words per sentences.
6. Time for talk is always limited hence select one idea at a time never include maximum technical information in the script.
7. Arrange the information in the sequences.
8. Include supporting and illustrated facts.
 

TV Talk
The talk delivered through TV i.e. through an electronic audio-visual medium which provides pictures with synchronized sound. The medium is cosmopolite in nature and can be used to develop instant mass awareness.

Important points while writing a programme for broadcasting
• Writing should be catchy, readable, conversational sound, understood and absorbed by the audience.
• Writing should be in simple, known language avoiding too much technicalities
• Interest of the listener should be sustained continuously
• Application of figures should be done judiciously
 

Principle of construction of flash cards
• Flash cards are non-projected visual aid in which series of illustrated cards are flashed as presented in a proper sequence to explain complete story.
• The story must be familiar, with specific heading and suggestions at the last.
• Simple words and local expressions should be used in flash cards.
• Total number of flash cards must be 10 -12 which are easy to remember to farmers
• Card: Select white or light coloured thick paper.
• Flash card should be large enough for everyone to see at least 22 x 28 inches and simple line drawing or photographs or cartoons having plenty of colours.

• Circular letter is a quick and cheap method of communication.

1. Attempt will be made to hold at Gram Panchayat level.
2. Before 2-3 days monadi of camps must be do.
3. The camp will be held for 1 day. It will start by 7 AM and close at 2 PM.
4. The venue should be selected so that it is in the central location to get cases from 3-4 villages in order to provide facilities of clinical examination, pathological tests, etc.
5. The venue should be centrally located and easily accessible to the general public.

The forest cover in most tropical countries has been under severe pressure and is diminishing at an alarming speed; consequently, they are experiencing severe biodiversity losses, reverting to a mature state at a slower rate with low resiliency. These tropical countries are aware of the danger causing due to the loss of forests, and large afforestation programmes have been launched by forest departments, private companies, and donor-supported projects. Nevertheless, plantation activities have rarely been able to keep pace with the deforestation rate, not to mention replacing what has been lost during the past. Plantation programme through afforestation activity is not only the matter of the forest department alone but also the combined responsibility of all the foresters. The appeal of forest planting has changed over the years following the broader demand spectrum.

To some extent, the forest plantations fail to create the diversity of plants and wildlife that has been lost. Hitherto, large-scale plantations are still necessary both directly to cater to the needs of industrial wood and help to minimize the pressure on the remaining natural forests. Types of tax system

1. Proportional tax: No matter the tax base’s relation to income size, the tax rate is always the same proportion. Taxing is according to the proportion of income. In this system poor people will be hit hard.
2. Progressive tax: Distribution of tax in a more just manner. People with higher income are charged with higher rates of tax. Eg. Income tax.
3. Regressive tax: Burden of payment will be more on poor. Taxes are charged from the poor at a higher rate than from rich. Ex. Commodity taxes – Affects poor rather than rich.

These hydrolysates are dried conversion products available in a powdered form. Up to 20 DE they are normally referred to as malto-dextrins or hydrolised cereal solids. Above 20 DE they are more commonly termed spray dried glucose syrup solids.

Applications

Spray dried hydrolysates find application in a wide range of processed and convenience foods. Principally as filler/carrier and sweetness reducer.

They are ideally suited for blending into powdered formulations, and serve the following purposes.

(1) Reduce loss of volume in contents on storage or in transit;

(2) Absorb fats and oils to retain free-flowing properties;

These products were originally flavoured with the herb marshmallow but a modem marshmallow is a foam that has been stabilised by a gelling agent. The traditional material for marshmallows is egg albumen which acts both as a foaming and gelling agent - this gives light, soft gel. Gelatine is now more commonly used, and once again can act as both a whipping and gelling agent. A gelatine marshmallow is soft and rubbery but heavier than an egg albumen one. Further gelling agents, e.g., pectin, agar and starch all give a soft, short textured gel. These gelling agents have the advantage of being vegetable products but have the disadvantage of normally needing a whipping agent as well. Enzyme-modified soya products have also recently become available for this use.

Methods of Marshmallow Manufacturing:

There are many bacteria and fungi which produce biofilms. These are made up of living, reproducing microorganisms. These microbes form communities and adhere tightly to the surfaces and are resistant to many antibiotics. Many pathogenic microorganisms form biofilms which are difficult to be removed and are harmful.Such microbial colonies produce an extracellular polymeric substance (EPS) to protect them. The dental plaque was the first documented evidence of microbial biofilms. When such microorganisms get a moist environment they attach themselves to any object and reproduce forming an attachment by secreting a slimy, glue-like substance. It is possible with metals, plastics, natural materials, medical implants, kitchen counters, contact lenses, walls of a hot tub or swimming pool. Initially it was the investigation of many scientists to show the presence of biofilms

Beneficial effects of biofilms have also been shown (Rosche et al., 2009) especially in agriculture (Bogino et al., 2013; Berlanga and Guerrero, 2016) where it can be utilized as a biological control agents. They are used as biofertilizers to enhance crop production (Timmusk et al., 2017), hazardous pollutants can be removed (Irankhah et al., 2019). 

Biofilms are resistant to antibiotics even at high concentrations (Costerton et al., 1999).This could happen when the diffusion of the antibiotic within the biofilm matrix is inefficient and results in slow growth rate of biofilm cells (Davey et al., 2000), there could be the presence of persistent cells and antibiotic-resistant small-colonies (Hogan and Kolter, 2002; Kolenbrander, 2000).Studies have proved that antibiotics can inhibit biofilm formation. An antibiotic azithromycin, is known to inhibit Pseudomonas aeruginosa biofilm. Biofilm colonies become resistant at higher concentrations of the antibiotics whereas at low concentrations these can be induced (Mah and OToole, 2001). P.aeruginosa is commonly present in the lungs of patients infested with cystic fibrosis. The clinical isolates show resistance to azithromycin (MIC>64 pg/ ml).However, low doses of azithromycin have shown improvement in lung function in patients with cystic fibrosis (Borriello et al., 2004). If antibiotics are given at low concentrations (below the MIC) than it induces biofilm formation in a variety of bacterial species. Sub-MIC concentrations of tobramycin can induce biofilm formation in P. aeruginosa. The use of antibiotics in agriculture, as growth promoters have shown to expose bacteria to low levels of the drugs and has caused havoc (Smith et al., 2002). Rachid et al. 

Acinetobacter baumannii is an opportunistic pathogen giving rise to nosocomial infections causing threat to public health. Studies of biofilm-forming genes was done in A. baumannii isolates by PCR technique. To study the relationships of genes a technique based on PCR sequences along with multilocus sequence typing (MLST) were utilized (Saeed et al., 2020). Goudarzi, Mehdi et al. (2019) studied the genetic diversity and biofilm formation analysis of Staphylococcus aureus causing urinary tract infections in Tehran, Iran. The biofilm forming and adhesion encoding genes were studied along with distribution of different agr and spa types in S. aureus isolates by PCR screening and multiplex PCR. The workers found that the ability to produce biofilm was higher among MRSA strains as compared to MSSA strains. The most prevalent biofilm forming gene was icaD (77.3%), followed by icaA (76%), icaB (57.3%) and icaC (50.7%). Acinetobacter baumannii is a nosocomial pathogen which is commonly present in hospitals and is a causal organism of many diseases. A. baumannii isolates have been known as multidrug-resistant (MDR). For controlling such serious organism only molecular techniques have been found to be effective. 

With recent work it has become clear that bacterial societies are very complex. Gene expression in bacterial communities are heterogenous. The heterogeneous gene expression contributes towards the biofilms in fluctuating environments which needs to be understood.Technique like single-cell analysis can help in the investigation of more complex communities. Recently, Jahid and Ha, (2012) have written a review on microbial biofilms of produce and the future challenges to food safety many outbreaks have been observed involving food- borne pathogens. Therefore, there is an urdent need for the safety of foods especially against microbial attacks and biofilm formation as they cause a risk to public health. Medically, the pathogenic microorganisms form biofilm- associated with the epithelial or endothelial lining which are embedded in the lungs, intestinal or vaginal mucus layer, the teeth or other medical implant surfaces or formed intracellularly (Dewasthale et al., 2018). Teichoic acids are characteristic major components of the cell surface in Gram-positive bacteria.

Cataract is a common cause of visual impairment and blindness in dogs and has a wide variety of aetiologies, as well as the disabling effect on the animal. Cataracts present important welfare considerations for the owner. Visual impairment in dogs may lead to increased nervousness, aggression and reluctance to exercise, all of which may adversely affect the owner-pet relationship (1). The only effective mean of treatment for cataract is surgical extraction of diseased lens (2).

Although vision is one of the important faculties for any living individual, dogs have very well developed olfactory lobe and have great sense of smell. They can move about easily in limited area familiar to them even with impaired vision. Cataract surgery was therefore not a common procedure in animals for many years. Quite often however,

All animals with cataract are not suitable candidates for surgery. Pre-surgical assessment of a candidate is essential to make a safe judgement for surgical procedure to perform lensectomy. Failure in canine cataract surgery can be associated with poor case selection. A thorough ocular and physical examination should be performed to rule out other ocular diseases prior to cataract surgery. The cataractous lenses in both eyes must be mature to cause functional loss of vision. (24).

The following prerequisites should be fulfilled before cataract extraction is recommended (6):

1. The affected eye should have a significant visual deficit.
2. For the patient to regain vision, its retina must be healthy and functional. if the fundus cannot be examined thoroughly (because of the cataract), retinal function should be evaluated with electroretinography (ERG) to ensure that retinal degeneration is not present as the patient history, signalment, and the speed of pupil contraction

Intra-operative complications

Although the phacoemulsification technique costs higher, it provides shorter surgical time and less complication than extracapsular lens extraction (49).

The most important intraoperative complications are anterior capsular fibrosis, radial tear of anterior capsule, posterior capsular rupture, vitreous prolapse and post-operative complications like uveitis and corneal oedema (50).

The production of ultrasound energy in phacoemulsification is associated with heat generation that can result in damage to ocular tissue, in particular the corneoscleral wound site. Thermal damage to the corneo-scleral wound site may result in difficulty with wound closure and consequent risk of wound leakage, as well as damage to the adjacent corneal stroma and endothelium, fistula formation, and the induction of high degrees of post-operative astigmatism. The loss of adequate flow of irrigation fluid around the phacoemulsification tip is the key factor in the development of phacoemulsification-induced thermal injury.

Introduction

A haemocytometer is a single piece of glass with an H-shaped trough forming two counting areas. Hemocytometers are used to determine the total cell count and viability of many different cell types The full grid on a hemocytometer contains nine squares, each of which is 1 mm¹. Each counting area is defined by a grid system. It has supports that hold the cover glass at the proper distance (0.1 mm) above these areas for the calculation of the concentration per volume of liquid.

Introduction

Most of the bacteria, excepting the photosynthetic bacteria, are damaged by ultraviolet (UV) radiation. UV rays have lower energy content and are capable of producing lethal effect in cells exposed to wavelengths in the range of 200 nm to 300 nm. Nucleic acids absorb these radiations leading to formation of thymine dimers which distort the structure of DNA. Distortion in DNA brings the interferences in DNA replication and transcription. The germicidal effect of UV can be used to determine the minimum amount of exposure required to produce 100% mortality of micro-organisms.

Introduction

The most important factor that influences the activity of bacterial enzymes is temperature. Most ofthe organisms grow under mesophilic conditions (at25⁰C). If the optimum temperature is increased, the rate of growth of organism is affected due to changes in protein structure which results denaturation of critical cell protein and ultimately inhibitsthe growth. If temperature is below critical temperature, then there is complete stoppage of growth as less activation energy is available for critical process to occur. However, there are certain organisms which can tolerate extreme temperatures viz.

1. Psychrophiles: Tolerate and grow at > 20⁰C temperature
2. Mesophiles: Tolerate and grow at between 20⁰C and 45⁰C temperature
3. Thermophiles: Tolerate and grow at <45⁰C temperature.

Introduction

The water potential plays an important role on growth of bacteria. If the medium has higher concentration of solute, it is said to be hypertonic. Due to this, the cytoplasm dehydrates and shrinks, resulting in inhibition of growth of bacteria. If solute concentration is less, the medium is hypotonic and hence maximum osmotic pressure on the cell.

Demonstration of biological nitrogen fixation is usually accomplished by estimating the total nitrogen through use of some modification of the Kjeldahl procedure. This technique is accurate and reliable whenever a medium initially low in nitrogen is employed.

Principle

The dried organism is digested with sulphuric acid. Organic and nitrate nitrogen is converted to ammonium sulphate and the ammonia gas is distilled into boric acid forming ammonium borate making weak boric acid alkaline. The ammonium borate is back titrated with standard sulphuric acid until the reaction mixture changes its colour from green to pink due to formation of ammonium sulphate. The standard acid used in back titration of ammonium borate is a measure of nitrogen content of the organism.

Introduction

Resazurin has high tinctorial value (related to colour), imparting characteristic blue colour. Resazurin test gives us indirect information concerning bacterial content and /or activity. Resazurin is also sensitive to the reducing action of leucocytes. The results of this test are comparatively less affected by antibiotic residues in milk than methylene blue reduction test.

Principle

Reduction of dye is indicated by a gradual colour change through various shades of purple and mauve to full pink, at which point the resazurin completely changes to resorufin.

Mammary tumors are either of benign or malignant types. Benign tumors are defined as mass or lump of cells unable to invade the neighboring tissue/ organ and less likely to re-grow after removal. However, malignant tumors are the tumors which have the ability to invade other tissues/ organs and tend to re-grow even after surgical removal. Malignant tumors grow at faster rate as compared to benign tumors. Nearly, 40-50% of canine mammary tumors (CMTs) are malignant.

CMTs can be classified on the basis of morphologic features, tissue of origin (epithelial, myoepithelial or mesenchymal tissue) or prognosis. The World Health Organization (WHO) classification of canine and feline mammary tumors has categorized CMT into three major categories i) malignant, ii) benign and iii) unclassified tumors. On the basis of histological characteristics, the mammary tumors have been classified into fibroadenomas, adenomas, solid carcinomas (with transformed epithelial cells), tubular adenocarcinomas (with tubule like arrangements), papillary adenocarcinomas (having papillary like projections), anaplastic carcinomas (with highly pleomorphic cells), sarcomas (involvement of connective tissue) and malignant tumors (Fig. 2). Fibroadenomas and adenomas are the types of benign mammary tumors the former contains some connective tissues component in between, and carcinomas, adenocarcinomas and sarcomas come under malignant tumors.

Human breast cancer (HBC) is a common malady concomitant with a massive death rate throughout the world. It has been reported that around 1.7 million women are globally identified with breast neoplasia, and out of this, half a million female dies from this terrible disease (Torre et al., 2015). Due to the shocking impacts of breast cancer, prevention, and therapeutic interventions have become a priority-based objective for many researchers to understand the biology of breast cancer disease during its progression and development.

The incidence of mammary tumor accounts for 25% of all the recorded tumors in dogs worldwide. Overall, about 198 cases of CMT are recorded per one lakh female dogs annually throughout the world which is quite an alarming percentage (Gray et al., 2020). Malignancy has also been recorded up to 45% which increases the fatality among morbid cases. Epidemiology of CMT is poorly documented particularly in India. In an epidemiological study conducted on canine tumors in Jammu in 2018, the overall prevalence was only 2% over a period of one year. The malignant tumors accounted 41.66% of the mammary tumors (Sharma et al., 2018). The rate of benign and malignant tumors incidence was reported as 46% and 54%, respectively. In another study conducted on the incidence of CMT in Ludhiana (Punjab) in 2020, it was found that 33.96% tumors were benign and 66.03% were malignant (Dash et al., 2020).

There are countless biomarkers present in cells related to and determining the progress of a disease. Identifying these markers could aid in detecting cells with metastatic potential (Ramaswamy et al., 2003).

The elevation of cyclo-oxygenase (Cox-2) expression was observed in CMT cases through enzyme immunoassay (Queiroga et al., 2005). In further study, enhanced expression of BRAC1and BRAC2 genes, certain enzymes such as matrix metalloproteinases (MMP-2 and MMP-9) by immunohistochemical examinations, and Bcl-2 & Bcl-XL elevated level in dogs with mammary neoplasm have been reported by Queiroga et al. (2011b).

A thorough history with complete physical examination including careful palpation of the mammary glands should be performed. As there are chances that dogs may be suffering from any other systemic ailments during middle to old age. In dogs with multiple tumors, all tumors should be excised and their location and size should be noted. Histopathological examination of the excised tumors should be carried out to classify mammary gland tumors. Mammary tumors are classified based on some of the parameters i.e., tumor size, lymph nodes, metastasis. Therefore, to stage a dog with mammary tumors information of these parameters needs to be collected and recorded. If lymph nodes are palpable and/or clinically enlarged, fine-needle aspiration should be performed for cytological evaluation. The technique is less invasive but with higher accuracy. Thoracic radiographs remain the mainstay for diagnosing the metastatic condition. The mammary tumors most commonly metastasize into lungs in dogs. The staging of CMTs becomes necessary as it is likely to confer some prognostic significance and provide guidance to veterinarians to make treatment decisions.

Surgical removal is the routine treatment method for mammary tumors, however, there is no consensus on the optimal procedure. There are supporting factors for every surgical procedure conducted for mammary tumors including lumpectomy (removal of tumor only), mastectomy (affected gland removal only), radical mastectomy (complete mammary gland and associated lymphatic tissue removal) and modified radical mastectomy (removal of complete mammary gland, lymph nodes and associated chest muscles).The more extensive procedures do not increase survival in dogs when compared to the other procedures, and the benefits of the simpler procedures are clear. Further, the radical mastectomy increases the disease-free interval but not the survival duration in cats.

One of the most promising areas of veterinary research these days is cancer immunotherapy and the development of CMT vaccines. DNA vaccines, also known as gene vaccines, are designed to transfer functional genes to target cells, allowing them to express functional proteins. Because endonucleases may easily access bare DNA, it requires a reliable and safe delivery route, which might be biological or non-biological. Different delivery strategies have been tried in human breast tumors to transport tumor antigens such Mucin-1 (MUC-1), Twist, HER-2, mammaglobin-1 and p53, as well as immunostimulatory molecules like IL-12 and IL-6. There have been various studies/ trials in canine animals using different molecules like IL-12, HER-2 and IFN-γ as vaccine candidates, with no clinically relevant outcomes or longlasting cell mediated and humoral immune response outcomes

In GADVASU, under the DBT sponsored project on “Development and evaluation of serodiagnostic assay for timely diagnosis and prognosis of mammary tumors”, we have generated and used hyper-immune sera raised against certain canine recombinant mammary tumor associated proteins for the standardization of sandwich and indirect ELISA for serodiagnosis of canine mammary tumor. The sensitivity and specificities of the assay’s ranges between 85-88% and 80-85%, respectively. However, an antibody against a single marker when used for different histotypes of CMT, its specificity reduced significantly. Multiplex assays are capable to detect many specific antigens in a single run whereas a traditional ELISA is used only to detect a single antigen. It has high-throughput potential, providing more results per sample, with lower sample volumes and with utmost sensitivity and specificity. Attempts are being made to increase the sensitivity and specificities of the assays by multiplexing various markers together (Fig.4).

Mammary tumors are the most common type of neoplasia found in female dogs with high percentage of malignancy and rate of mortality. The rate of incidence of these mammary neoplasia and dysplasia is continuously advancing nationally and internationally over the last 45 years. There are different risk factors such as age, diet, hormonal exposure and breeds, which are required to be monitored in order to control the incidence of CMTs. In addition to conventional therapies such as surgical excision, radiotherapy and chemotherapy, additional precision therapeutic agents targeting TKIs, p53 tumor suppressor gene, cyclooxygenases, epithelial-mesenchymal transitions and various hormones/ hormonal receptors could be beneficial for the treatment of CMTs. Scientific efforts are also being made to develop different biomarker panels and tests for prognosis/diagnosis of the diseases as well as to develop vaccines against CMTs.
