Introduction

Wet ashing is suitable for the determination of Ca, Cu, Fe, Mg, Mn, K, Na, Se and Zn in biological samples and may be applicable to the determination of other elements as well.

The organic matter of the sample is oxidized with concentrated nitric acid and perchloric acids. The acids are partially removed by volatilization and the solube mineral constituents remain dissolved in nitric acid. Any silica present is dehydrated and made insoluble.

Estimation of Cellulose in Feeds and Fodders
Principle

The feed well ground is digested with acetic acid which dissolves all other constituents like protein, disaccharides, fatty acids, etc., and cellulose and mineral contents are unaffected. The residue is made acid and alcohol free by washings. Alcohol removes water, benzene removes pigments and fat and at the end the sample is dried and ashed. The difference between the dried weight before ashing and weight after ashing gives cellulose content of the feed sample.

Method I
Reagents
    1.    20% sodium carbonate.
    2.    Conc. hydrochloric acid.
    3.    4% sodium hydroxide
    4.    10% calcium chloride
    5.    Ammonium hydroxide
    6.    Potassium permanganate (0.05 N)

Procedure
Take 2-5 g of powdered sample in a beaker without spout, and add 40 ml of water and boil for half an hour. In case of faeces, add 40 ml of 4 percent NaOH instead of 40 ml of water to form silicates. Now boil, for half an hour with cold water flask over the beaker, then add 10 ml of 20 percent sodium carbonate solution and boil for  another half an hour. During these two cooking processes, the following changes take place.

Principle

Cyanide is distilled from chloroform and water soultion into KOH solution, forming potassium cyanide. KCN is titrated with silver nitrate; 2 KCN + AgNO₃ = KCN. AgCN + KNO₃. An excess of AgNO₃ produces insoluble AgCN, which is the end point of titration.

Principle
(Method of Patterson 1963)

The albumin bound long-chain fatty acids of plasma are usually referred to as unesterified or non-esterified fatty acids (NEFA) to distinguish them from the long-chain fatty acids present in the esterified form in neutral fats and phospholipid. In the case of ruminant plasma, it is also important to distinguish these “free fatty acids” from the small molecular weight steam volatile fatty acids of dietary origin which are found in the free state as the carboxylate ion.

Basically, the method of Dole (1959a) has been used to estimate plasma NEFA employed by Annison (1960), which is to evaporate the heptane extract of plasma to dryness before titration. This is conveniently done in a stream of nitrogen on a warm hot plate rather than by evaporation in vacua. In place of the 0.05 M- phosphate buffer (pH 6.0) of Annison or the 0.02 N-H₂SO₄ used by Dole (1956a), in the present method 0.02 N-HCI is used as an extraction medium. This is preferred because owing to its volatility, it is less likely to cause serious accidental contamination of the supernatant heptane layer.

Principle

A simple and rapid enzymatic method for the quantitative estimation of total cis-methylene- interrupted polyenoic acids has been devised. Linoleic, linolenic and arachidonic acids, the more common acids of this group were used to calibrate the method. The potassium salts of the fatty acids are oxidised by atmospheric oxygen in the presence of the enzyme lipo-oxidase, and the absorption of the conjugated diene hydroperoxide is measured at 234 mµ. As little as 5 µg of linoleic acid can be quantitatively measured with good accuracy and precision. The total content of polyunsaturated fatty acids containing the cismethylene-interrupted diene structure of fats, oils, hydrogenated oils, fatty acids, esters blood plasma, microorganisms and plant seeds has been measured directly by this method.

Gravimetric Method — Principle

Total lipids in plasma consist of neutral fats, phospholipids, chloesterol esters and free cholesterol. Minor components include non-esterified fatty acids (NEFA), phosphatides and sphingo myelin. Total lipids in serum or plasma are extracted using ethyl alcohol and petroleum  ether, separated and weighed after drying at 105ºC.

Procedure
Pipette 5 ml serum in a separating funnel, add 5 ml ethyl alcohol and 12.5 ml pertroleum ether. Shake the contents of funnel two times for 3 min. atleast. Keep the contents of funnel as such for one hour. Transfer the contents of funnel in the moisture cup, dry and weigh. Calculate the result by difference method using the following formula.

Introduction

(Titrimetric Method of  Cherry and crandall, 1932 Modified by Tietz and Fiereck, 1966).

In 1932, Cherry and Crandall employed olive oil as the substrate for pancreatic and serum lipase (LPS. Triacylglycerol lipase) determinations. In subsequent years this method was improved (Tietz et al., 1959; Tietz and Fereck, 1966: Tietz. 1972), reducing the original 24 h, period to 6 h and finally to 3 h.

Principle
This procedure is based on the hydrolysis of triglycerides in olive oil into fatty acids, diglycerides and to some small extent into monoglycerides and glycerol.

Investigation of the effect of Various Hormones on Fat Synthesis from 14C Labelled Acetate in Chicken Adipose Tissue

(Method of OMea and Leveille, 1968, Folch, Less and Solane Stanley, 1957 and Vaughan, 1961)

Reagents
    1.    Krebs-Ringer calcium-free bicarbonate buffer 7.4
        100 parts 0.154M NaCI (0.9001 g in 100ml)
        4 parts 0.154M KCI (0.0459 g in 4 ml)
        1 part 0.154M  MgSO₄ (0.0380 g in 1 ml)
        (3 parts 0.110M CaCI₂)
        21 parts 0.100M NaHCO3 buffered to 7.41 by bubbling through CO₂

Hypothesis

The validity of measuring urea kinetics using a single dose of ¹⁴C-urea and using urinary excretion as a sampling system is based on specific assumption:

    1.    There is a rapid equilibration of ¹⁴C-urea with the endogenous urea pool following infusion. This pool is conceived to be a measure of all body compartments permeable to urea and in equilibrium with the plasma urea pool. 

    2.    Experimental animals are metabolically maintained in a steady state during the test period and metabolic breakdown of urea is random.

    3.    Specific radioactivity for urinary urea is proportional to the dilution of replenishment and simultaneous depletion rate of the urea pool.

    4.    That the metabolic parameters of urea are correctly measured.

Pepsin Digestibility of Food
Principle

Solvent extracted samples are digested for 16 hour with warm acid solution of pepsin under constant agitation. Insoluble residue is centrifuged, dried and weighed and analysed for protein ; or filtered, washed and analysed for protein.

Apparatus
    1.    Centrifuge : Capable of at least 1750 rpm with conical bottom tubes of 150 ml capacity. If necessary, solution may be centrifuged in 50 ml tubes.
    2.    Agitator : or water bath with shaking device and having temperature controlling unit.
    3.    Glass fiber filter paper.

PEG Estimation
Introduction
Polyethylene glycol (PEG) is used as a marker for the estimation of rumen water volume and ruminant digestion studies (Hyden and Ekman, 1953). Long back, Hyden (1955) developed a turbidimetric method which is described below :

Principle
When trichloroacetic acid is added to a solution of PEG, a turbidity is produced, which can be measured photo-electrically. The intensity of the cloud, which consists of finely divided oily droplets, is increased in the presence of barium ions. Protein and sulphate must be completely removed from the sample before the addition of the trichloroacetic acid reagent. Sulphate is precipitated with barium chloride and the proteins are removed essentially according to Samogyi (1945). The method has been developed for determination of PEG in ruminal and abomasal fluids, but it has been tried with other biological materials too.

Enzymes are biocatalysts composed of amino acids. Enzyme reactions are highly specific. It binds to the substrate to form a reversible intermediate product called enzyme substrate complex. As the concentration of the substrate increases the rate of formation of the intermediate complex also increases. This increase in velocity of the reaction is in a hyperbolic manner till it achieves the maximum velocity called Vmax .At this point all the enzymes will be in the form of ES complex. Substrate concentration will be in at its optimum level i.e. further increase in substrate concentration will not affect the rate of the reaction. The substrate concentration at which the rate of the reaction is half the maximum velocity Vmax is called the Michaelis-Menten constant, Km. This is constant for the given enzyme acting on the same substrate.

The cervical vertebrae are 7 in number.

The first and second cervical vertebrae are highly modified in conformity with the special function of support and movements of the head.

The third, fourth and fifth have the following characters:

The body is long. Ventral surface present a median ventral spine.

The dorsal surface has a flat central area which is narrow in the middle of the vertebrae, and wide at either end. On either side of this area there is a groove which lodges the longitudinal spinal vein.

The anterior extremity or head has an oval articular surface which faces forward and downward; it is strongly convex and wider above than below.

The posterior extremity is larger and has a nearly circular cotyloid cavity.

The arch is large and strong. It is perforated on either side by foramen which communicates with the foramen transverse hum. The vertebral notches are large.

The articular process are large. Their articular surface are extensive, oval in outline, and slightly concave.

LUMBAR VERTEBRAE (Fig. 1)

Lumbar vertebrae, six in number, are much longer than in the horse.

The body is much constricted in the middle, expanded at either end, and bears a rudimentary ventral crest.

The articular processes are large, and their facets are more strongly cun/ ed than in the horse.

The transeverse processes ail’curve foPA/ard, borders a.^e thin and irregular and often bear projections of variable size and form..

The spinous processes are relatively low wide, the last being the smallest; their summits are moderately thickened.

DIGITS

Four digits are presents in the ox of these two the third and forth -are fully developed and have three phalanges and three sesamoid each.The second and fifth are vestiges.

FRIST PHALANX- (Fig.No.1)

The interdigital surface is flattened and its volor parts bears a prominence for the attachment of the interdigital ligaments.

The proximal extremity is relatively large and somewhat compressed from side to side.

The articular surface is concave from before backword and is divided by sagittal groove in two areas, of which the aboxial one is the larger and higher.

Behind tese are two facets for articulation with the sesamoid bones.

The distal extremity smaller than the proximal.

First Aid in Orthopaedic Patient

Immediately transfer the animal to a safety place.

The place should be well settled, well ventilated, neat and clean.

If the fracture site is limb and it is a simple fracture, immediately immobilize the animal with soft cotton padding and plastic scale placed at four sites.

If the fracture site is limb and it is a compound fracture, cover the fractured site v/ith th’ck padded clean cotton cloth and few bamboo splint placed on four sides to keep limb straight.

Control the bleeding, if bleeding occurs at time of covering of fractured site with thick padded clean cotton cloth.

If the fracture site is vertebral column, it is better not to lift the patient vigorously.

A patient with vertebral column fracture shouldn’t be allowed for standing.

Aim

Definition

Fracture is a disolution of bony continuity with or without displacement of the fragments.

Indication -To know the method of POP bandaging technique.

Method of preparation of POP bandage rolls-

A gauze bandage of 6" or 4" width is spread on a clean floor. Plaster of paris powder is spread uniformly over the entire length of bandage & is folded into roll bandage. In this way 4-6 bandage rolls are prepared.fig-1(a)&(b)

Method of preparation of pop bandage splints

The length of the prepared pop splint is determined by measuring the entire length of fractured bone along with the joint below and above the fractured site) are measured. This much of length of gauze is spread for splint preparation.

POP powder is impregnated upon gauze. The gauze is again spread over this much of length in this manner sprinkling of POP powder & overlapping gauze bandage upto 4-6 layer are continued [fig-1 (b)]

Aim: To know the technique of application of first layer padded cloth and its thread

DEFINITION

Lower level fracture in animals include any fracture of the region from knee joint to foot in forelimb and from hock joint to foot in hind iimbs.

CFERA’nVE PROCEOURE-H ere the principle of treatmient is application of cast and conjoint metal ‘U’ plate over the fractured site. Permanent immobilization of the limb should be done only after 7 days to subside.

Length to be covered-Forelimb-above knee joint and below foot. Hind limb-above hock joint and below foot, (as shown in fig.1)

Padding-6-12 layer thick padded cloth should be vmapped applying gum.s throughout the doth area, over the fracture, it should not be too tight but it should be strong. Sloughing off the hoof may result due to hypostatic congestion; hence the hooves should not be left open. (Cotton pad as shown in fig.2)

Definition

Fracture of radius-ulna & tibia-fibula comes under middle level fracture.

Methods of Fracture Repair

External coagtation bandage along with thomas splint application.

Technique for Fracture Repair

Animal controlled with lateral recumbency with fractured limb upward.

Definition

Fracture of humerus and femeur bone has been grouped under higher level fracture. Pinning as well as Thomas splint application are considered to be the approved method of immobilization for repair of these bone fracture.

Both method of repair are highly essential. Any one method of repair has been considered dangerous for the animal. Only pinning of the bone makes the limb to be turked where as only Thomas splint application disturbes the bone alignment at fracture site. Hence leaving one method of repair other method is not possible.

When above method of repair is not possible a modification of velpue sling can be attempted.

Indication: Highest level fracture (femur fracture) (Fig. 1)

Aim:- To know the measurement for preparation of an appropriate size of ring of Thomas splint for a large animal.

Indicaion

To know the importance and method of net fabrication in large animals thomas splint. Objective;

To make the application of thomas splint lightest.

To avoid use of plaser of pahs bandage.

To prevent possible all direction of the immobilised limb in thomas splint.

Aim: To secure the foot on the distal end of Thomas splint.

Procedure

1. Foot is placed upon the distal end of Thomas splint.
2. The foot of the animal is wrapped V’- two round with bandage cloth.(Fig.1)
3. 2^nd round of bandage cloth is wrapped around the distal end of Thomas splint.(Fig.2)
4. In 3^rd attempt, the bandage roll is taken from the ground level of the foot & including the distal end of foot.(Fig.3)
5. The bandage roll is taken through the gap between anterior end of Thomas splint.(Fig.3)
6. In 4th attempt, the bandage roll is taken from the volar or plantar aspect of the limb and passed upon the ground level of foot as well as the distal end of Thomas splint.(Fig.4)
7. Final 2 rounds of bandage cloth is wrapped through entire surface of foot.

Definition

Proper healing of amputed stump, avoiding infections.

Description

Limb is amputed generally through diaphysis or by disarticulation method. Whatever is the amputation,the surgeon should always keep as much soft tissue as possible to cover the bone extrimities. It is difficult to achieve in large animal, when amputation is immediately below carpus or tarsus.

Joint disarticulation: Unwanted limb is amputed by joint disarticulation method.

1. The plaster-bandage wrap cast :(Negative cast)

The amputee was restrained on lateral recumbency keeping the amputed leg upwards.

A thin, moistened stockinet was pulled over the stump and it was maintained snugly in place by tying with a string at the top.

The moistened cast stockinet was marked with indeliable pencil indicating prominences with a cross and other sensitive areas with a circle (Fig.1).

Plaster bandages were then wrapped as pen requirement unit I the shell has a thickness of approximately 3mm in proximal third.

After plaster hardened, it was removed either by pulling out or longitudinal splitting with a cutter (Fig.2).

Introduction

In India there are about 200 distillery having installed capacity 1555.5 million liters per annum out of which 50% are of 30 KL/ per day capacity about 50% of remaining distillery are of 45 to 60 KL/ per day capacity there are few distilleries which having go 120 KL/ per day capacity. Distillery waste is one of the major industrial waste having high organic and inorganic constituents in the recent past the treatment and disposal of distillery waste has become a subject of debate of each and every forum.
