If the 1990s were the decade of genomics, the first ten years of the new century are set to become the decade of proteomics. For the first time, the technologies of proteomics make it possible to generate quantitative protein expression data on a scale and sensitivity comparable to that achieved at the genetic level. This advance has major implications for our understanding of cellular organisation in health and disease and for pharmaceutical and agricultural biotechnology. Indeed, proteomics is already yielding important findings, across a wide range of applications. Most proteomics research, however, is directed towards the more proximal goal of investigating protein expression and function under specified physiological conditions.

The rapid evolution of proteomics has continued during the past years with a series of innovations in the core technologies of two-dimensional electrophoresis and mass spectrometry, and a diversity of productive research programmes. Well annotated proteomics databases are now emerging in a number of fields to provide a platform for systematic research, with particularly promising progress in clinical applications such as cardiology and oncology. Large-scale quantitative research, comparable in power and sensitivity to that achieved for gene expression, is thus becoming a reality at the protein level.

Gene silencing is the suppression of gene expression through nucleotide sequence-specific interactions that are mediated by RNA. Many manifestations of RNA silencing-RNA interference (RNAi) in animals, quelling in fungi and post-transcriptional gene silencing (PTGS) in plants, appear to be related. RNA silencing is experimentally activated by double-stranded RNA (ds RNA) and is used as a powerful technique for specific inhibition of gene expression in a variety of organisms. In a natural context, ds RNA may be produced from rearranged loci by transcription from converging promoters or by host-or viral-encoded RNA-dependent RNA polymerases (RdRP). A key component of RNA silencing is 21-23 nt RNA known as short interfering RNA (Si RNA). In Drosophila, the siRNA is derived from ds RNA by the action of an RNase-III like enzyme named DICER. The siRNA guides a multi-subunit endnuclease, referred to as the RNA-induced silencing complex (RISC) and it specifically degrades RNA that share sequence similarly with the ds RNA.

Biotechnology provides significant opportunities and greater effects in horticulture, where even minor changes such as quality, colour, aroma, flavour, structure, disease-pest resistance and postharvest behaviour bring tremendous achievements commercially. Tissue culture, genetic engineering, molecular breeding, biofertilizers, biopesticides and postharvest biotechnology are important aspects of biotechnology in horticulture such as fruit, vegetable, ornamental, spice, medicinal, aromatic and plantation crops.

Tissue Culture

Tissue culture is the true reality of the plant biotechnology. The annual demand of tissue cultured production constitutes nearly 10 per cent of the total world biotech business, amounting to 15 billion US dollars. The global production of tissue culture industry is expanding @ 15% per annum (Chadha, 2000). In India, more than 130 tissue culture units are presently producing > 300 million plantlets per annum (Choudhury and Tejaswini, 2000).

The importance of Plant Pathology has been realized over the years owing to number of crop failures in different parts of the world. Plant disease control is indispensable for qualitative production and to compete in global agricultural trade. There is great need of sensitization of our research in various aspects which shall guide the generation to come for managing plant diseases not only for enhancing crop production but also in facilitating trade by conserving the biodiversity and environment.

Research Trend in Plant Pathology

Like in other sciences, there has been a change in the trend of research in Plant Pathology with passage of time. Up to the late 1960s most of the interest in plant pathology was concentrated in crop diseases mostly caused by fungi. Physiology of microorganisms and physiology of parasitism became increasingly popular fields of interest in 1960s.

The advancements in the field of informatics, biology and allied fields of technology led to the emergence of an inter-disciplinary field known as ‘Bioinformatics’. This field is an expanding and challenging tool for the collection, storage, organization, integration, analysis and simulation of biological data for utilization in biotechnology.

Collection of Data for Bioinformatics

An epoch making revolution in Genomics in recent years in deciphering plant, animal and microbial genome sequences, Proteomics, Statistics, Instrumental techniques, Structural and Computational Biology have given input to fragmented pieces-and-bits of information on biological materials. Each data is a discrete piece of important but defragmented information regarding a molecule (protein, RNA, DNA, sugars or lipids etc) or a reaction or at best a pathway, supposed to be occurring in a living system independently at a given time and space. However, as in a living system each molecule or a process exists in unison with other entities of its kind, these fragmented data are to be stored, organized, and then integrated and analysed to form an integrated part of a whole chain of events – now called a ‘SYSTEM’. Storage and organisation of data Explosion of knowledge in the field of biology and generation of new informations (data) needs well storage, organization and a faster distribution among the scientific workers. The advent of super computers, high speed computing and development of algorithms for data formatting, assists in the storage of data at the site of origin and distributed through the hyperlinks in world wide web (www).

Drug designing is the approach of finding drugs by design based on their biological targets. The substance used for medical purpose, either alone or in mixture that changes the state or function of cells, organs or organism as a whole is known as a drug. So typically a drug target is a key molecule involved in particular metabolic or signaling pathway that is specific to disease condition or pathology, or to the infectivity or survival of a microbial pathogen. Drugs may be designed that bind to the active region and inhibit the key molecule that causes disease. However these drugs would also have to be designed in such a way that they should not affect any other important molecules similar to the key molecule. These can also be used to enhance the normal pathway of metabolism by promoting specific molecules that may have been suppressed in disease state.

Bioinformatics can be defined as “the collection, archiving, organization and interpretation of biological data” (Orengo et al., 2003). By extension, structural bioinformatics deals with the collection, archiving, organization and interpretation of 3-D structural data of bio macromolecules. Structural bioinformatics emerged with the first crystal structures of haemoglobin and myoglobin. These structures were analyzed and contributed significantly to our understanding of the principles of life at a molecular level. A repository of all publicly accessible protein crystal structures, the Protein Data Bank (PDB) was established by Walter Hamilton et al., at Brookhaven National Laboratory in 1971. Until the late 1980s, structural biologists often knew all the available structures of bio macromolecules and much of the analysis, such as structure comparison and hydrogen-bonding analysis was done manually without much computer intervention. As the number of available structures increased, so did the need for software tools for structure analysis. Structural bioinformatics encompasses a wide variety of applications, including structure visualization, validation, classification, structure-based function prediction, identification of targets for structural genomics projects and prediction of the structures of individual bio macromolecules and complexes as well the binding modes of small molecules inside proteins of pharmaceutical or biotechnological interestStructural and theoretical analyses of proteins are central to the understanding of complex molecular mechanisms and are fundamental to the drug discovery process.

Biodiversity conservation and sustainable use has, for many people, become associated mainly with issues related to plants and wild animals. Although much less discussed, the loss of farm animal genetic resources may well be much more serious than in crops because the gene pool is smaller and very few wild relatives remain. The fact that 32% of livestock breeds worldwide are at risk of becoming extinct and that the rate of extinction continues to accelerate (FAO, 2000) is thus a serious cause for concern. Animals of different characteristics and hence outputs suit differing local community needs.

Among several respiratory diseases affecting man, bronchial asthma is the most common disabling syndrome, nearly 7 to 10 per cent of the world population suffer from bronchial asthma, which is a chronic illness involving the respiratory system in which the airway occasionally constricts, becomes inflamed, and is lined with excessive amounts of mucus, often in response to one or more triggers. These episodes may be triggered by such things as exposure to an environmental stimulant (or allergen), cold air, warm air, moist air, exercise or exertion, or emotional stress. In children, the most common triggers are viral illnesses such as those that cause the common cold. A variety of exogenous determinants of asthma were also been, some of which are air pollution, tobacco smoking, diet, occupation and respiratory infection, these pollutants may serve as inciters or triggers of asthmatic reaction in airways that are already hyper responsive, they may exert direct toxic influence on the respiratory epithelium or may augment or modify immune responses to inhaled antigens. During an asthma episode, inflamed airways react to environmental triggers such as smoke, dust, or pollen. The airways narrow and produce excess mucus, making it difficult to breathe. In essence, asthma is the result of an immune response in the bronchial airways. As according to the World Health Organization over 80% of the people in developing countries depend upon traditional medicine for their primary health care. Medicinal plants are considered as source of various alkaloids and other chemical substances essential for mankind.

A fundamental problem facing researchers today is how to make effective use of available wealth of online background domain knowledge to improve their understanding of complex biological system. Effective use of such online background domain knowledge plays a crucial role in all stages of integrated biological studies.

In the past years, research in molecular biology and molecular medicine has accumulated enormous amounts of data. This includes genomic sequences gathered by the Human Genome Project, gene expression data from micro array experiments, protein identification and quantification data from proteomics experiments, and SNP data from high-throughput SNP arrays. However, the understanding of the biological processes underlying these data lags behind. There is a strong interest in employing methods of knowledge discovery and data mining to generate models of biological systems. Mining biological databases put forward challenges which knowledge discovery and data mining have to address in the future. Important background knowledge in bioinformatics is often contained in documents, such as scientific publications or database annotations. Analyzing data from biological databases often requires the consideration of data from multiple relations rather than from one single table. Recently, approaches (such as propositionalization algorithms) are being studied that utilize multi-relational data and yet meet the efficiency requirements of large-scale data mining problems.

Algae refer to an assemblage of polyphyletic organism that conducts oxygen evolving photosynthesis other than land plants. They are distributed widely throughout India in the sea, in fresh water and in moist situations on land. Algae also form an important component of biodiversity of Andaman and Nicobar Group of Islands. Database for Algae resources of Andaman and Nicobar Islands are created with an aim of providing an information pool to the researchers working in fields related to fresh water and marine water algae in Andaman and Nicobar Group of Islands. About 313 fresh water algal species belonging to 15 families and 57 marine algal species belonging to 18 families has included in this database among which 40 species has been found to be first time reported in India. Algae have a wide application in a variety of Industries such as Food, Fodder, and Fertilizer etc. They are also important sources of Protein, Iodine, Vitamins, Minerals and Substances of Antibiotic Nature.

Data Mining is the process of automatic discovery of novel and understandable models and patterns from large amounts of data. Bioinformatics is the science of storing, analyzing and utilizing information from biological data such as sequences, molecules, gene expressions, and pathways. Development of novel data mining methods will play a fundamental role in understanding these rapidly expanding sources of biological data. Data mining techniques are an automated means of reducing the complexity of data in large bioinformatics databases and of discovering meaningful and useful patterns and relationships in data. Bioinformatics, the application of computational techniques to analyze the information associated with biomolecules on a large-scale, has now firmly established itself as a discipline in molecular biology, and encompasses a wide range of subject areas from structural biology, genomics to gene expression studies. It provides an introduction and overview of the current state of the field. The main principles that underpin bioinformatics analyses look at the Data Mining in Bioinformatics and databases that are commonly used, and finally examine some of the studies that are being conducted, particularly with reference to transcription regulatory systems.

Management of Compound Fracture

An open fracture is an injury that occurs when a broken bone is exposed through the skin. This may mean that the bone is actually sticking out of the skin, or it may mean that the skin and soft-tissue is disrupted and exposes a path to the site of the fracture. Often called a compound fracture, an open fracture necessitates different treatment from the usual closed fracture.

Surgically Cleaning the Bone

Surgically cleaning the bone is one of the first steps for treating an open fracture. Most patients who sustain an open fracture undergo a surgery called “irrigation and debridement.” Irrigation means washing the bone and the site of the injury. Debridement is described in the next step.

Determining the extent of injury can be difficult just by looking at an open fracture. This is especially true in high-energy injuries including automobile collisions and gunshot wounds. With these types of injuries, even small penetrations in the skin can cover very large areas of soft-tissue damage around an open fracture. Therefore, when surgically cleaning the bone, it is important to do this in the operating room (OR) under anesthesia — trying to sufficiently assess and clean the bone in the emergency room, without adequate anesthesia, may be insufficient.

Eastern India is frequently affected by weather hazards, such as drought, flood and heat wave. The region consists of Orissa, West Bengal, and Bihar. The coastal states of Orissa and West Bengal frequently face cyclones, while Bihar and eastern U.P. face cold wave. Due to unfavorable seasonal weather, crop productivity is poor. However, the recent change in climate induced by global warming has further threatened the agricultural productivity in the region. Rice is the principal crop of Eastern India and rice based cropping systems has long been practiced in the region. While rice-rice is the most dominant cropping system in Orissa and West Bengal, rice-wheat is in Bihar and eastern U.P. Besides, rice-groundnut occupies a considerable area in the region. It is now doubtful whether the resilience of these cropping systems would continue sustaining the productivity under the climate change. The major aim of the proposal was to identify suitable adaptation measures of crop production under projected climate change. Climate change is expected to alter precipitation pattern, increase temperature, change in humidity level and decrease sunshine. As a result it is like to influence right from land preparation to the harvest and post harvest process. Thus the project aim was to revisiting the existing package of practices of selected cropping systems by simulation modelling, deduce suitable adaptation measures and validate these by OTC experiments.

Object: Methods of measurement of water status and water saturation deficit in plant parts.

Theory: Plant water status strongly influences plant growth and biomass production through its effects on leaf and root expansion and on photosynthesis. Therefore, measurement of plant water status is an important component of understanding plant processes and maximizing yield under different ecosystems.

Theory: Yield is the ultimate objective of cultivating agricultural plants. Different plant parts constitute the economic yield in different crops, like grain in cereals, pulses and oil seeds, vegetative parts or fruits in record yield at the correct maturation stage. Do not allow excessive wilting or drying of the material. Estimate yield in a sufficiently large sample. Materials required only a representative crop to estimate yields. (Do not select plants of any particular type. Do not damage any plant part vegetable crops; bark or out growth in fiber crops (jute, cotton); and the entire plant in fodder crops. The yield per unit area (per M₂ or ha) is the product of yield per plant multiplied by the number of plants per unit area. It is, therefore, essential to know the yield components per plant as well as per unit area.

Water is the most important and abundantly required nutrient for poultry. A continuous supply of clean, fresh and wholesome potable water is essential for poultry. Poultry rearing is becoming more and more intensive with which the chances of the infectious diseases being sustained in the different flocks also increase. Consequently, the need for effective disease control measures to be adopted also gains further importance.

Most often, poultry farmers get alarmed only when the mortality level or death rate in a farm is high. However, even the existence of disease at a sub-clinical level may hinder the performance of the birds, in terms of body weight or egg number; such economic losses, sometimes relatively less and unnoticed, may mean the difference between success or failure in the poultry business. Treatment of a sick flock after the onset of any disease becomes costlier and more frequently impracticable. The recovered birds may not regain their production to original levels and may also continue as source of infection to other healthy birds. Hence, the adage ‘prevention is better than cure’ applies more to poultry industry than any other field.

Puberty and sexual maturity

Puberty is defined as the age at which reproductive function is initiated in animals. This corresponds to changes in hormone levels and the ability to function sexually in order to reproduce. The puberty is the period when the sexual organs are functionally developed, libido is prominent and the male is able to produce the spermatozoa and is able to copulate resulting in pregnancy. The onset of puberty is a gradual process and occurs after achieving optimum body weight, which is influenced by plane of nutrition, genetic differences, management practices, breed, diseases and individual differences.

Semen is admixture of secretions of testes containing spermatozoa and accessory sex glands. Spermatozoa are formed in the seminiferous tubules of the testes. The tubules contain a complex series of developing germ cells that ultimately form the male gametes. The spermatozoa are unique type of cells containing nucleus (head portion) and a tail. The acrosome is a double walled structure situated between the plasma membrane and the anterior portion of the sperm head and neck connects the sperm head with its tail.

1. AI with liquid semen

The ampoule is thawed in ice water for 6-7 minutes. Then, the ampoule is dried and cut. The semen is drawn in a sterilized glass pipette (40-42 cm x 5 mm length diameter) fitted to a sterile glass/plastic syringe (2-5 mL) filled with 1 mL of semen with a rubber connecter. Breeding soundness examination can be used as a diagnostic aid in herd fertility problems. After breeding soundness examination, a bull is classified as a satisfactory potential breeder, a questionable potential breeder or an unsatisfactory potential breeder. Members of the Society for Theriogenology developed the concept of a breeding soundness examination as mentioned below.

The first buffalo AI progeny is reported at Allahabad in August 1943. Although AI is commonly used as a tool of breeding in buffaloes and complaints have appended that conception rate is not satisfactory. There are differences in the anatomy of the sexual organs, sexual physiology and general breeding behaviour that have to be known and taken into consideration when the same technique of AI used in cattle is applied in buffalo breeding. It explains the lower semen production capacity of male buffaloes compared to the cattle bulls. The buffalo bull’s penis is shorter and thinner than penis of the cattle bull. There is a pronounced tapering of the penile end and glans penis is rather poorly developed. The thrust is shorter and less forceful than the bulls. The sheath is tighter and closer to the abdomen than zebu cattle. These anatomical differences are of significance in the semen collection work and type of artificial vagina to be used. The weight of testis and epididymidis in the male buffalo is only ¼ of what it is in cattle. In addition, the seminal vesicles and the prostate are considerably smaller.

Review of the state of art of Drought on impact assessment, monitoring and management with special reference to agricultural production has revealed that Drought is still a complex natural disaster and difficult to forewarn in spite of advancements in atmospheric sciences and computational procedures. It is realized only after it is set in and is not known when it is likely to withdraw. A long way one has to travel to predict its occurrence, spread and its continuation to the next season over a region. The existing net work of observational set up in developing countries in Africa, Asia and in South America need to be strengthened further for effective monitoring of significant weather events at regional and micro regional level. With the rapid expansion of communication system over the past two decades, establishment of Automatic Weather Stations (AWS) at as many locations as possible even in remote regions should be taken up on priority basis and funding may be provided by the respective governments or by the International Organizations where ever necessary. Nations with limited resources may also be convinced to strengthen their weather observational net work in view of the increased occurrences of climate extremes and changing climate. Drought vulnerability regions over the globe were delineated based on sparse historical weather data which do not match with real surface conditions. Now, with the availability of weather data from more number of stations both temporally and spatially, drought prone regions over the world may be re-delineated. This would help the planners to allocate funds rationally to the affected regions. Globally every year, some regions or others may be experiencing droughts and more so in drought prone regions.

ORIGIN OF INSECT

Entomology is a branch of zoology which deals with insects. In this branch we study the insect origin and evolution of insect and their diversity and classification and other parts of insect body. Insects belong to the class insecta in the phylum arthropoda, the largest group in the animal kingdom. All arthropods are characterised by having segmented body, claws, chitinous exoskeleton, ventral nervous system and dorsal heart. The class insecta is generally considered to have evolved from myriapod of some sort during the Devonian period. Based on different mandibles and mandibular movement concluded that insects are not direct insect group myriapods the mouth is located between the prostomium and the first body segment and the anus open in the periproct.
The first great step was the development of a pair of ventral appendage. Excretory system of an organism maintains its homeostasis i.e., a constant internal environment. Here in insects, the blood is haemolymph therefore it determines the excretion of an insect’s body. Excretion also eliminates the nitrogenous wastes and maintains the salt- water balance and also regulates the uniformily of haemolymph. The characteristic feature of insects is the presence of malpighian tubules to eliminate the metabolic wastes, whereas rectum walls carry out the function of reabsorption of salt and water. A minimum amount of water is excreted with nitrogen waste in order to conserve the water in body.

Locusts are injurious to man as it destroys several human commodities. Locusts are phytophagous and feed on several plants cultivated by man. The locusts are insects belonging to the order orthoptera. The most popular insect is grasshopper. The only difference between the two types of insects is that locust can exist in two different behavioural states whereas grasshoppers do not. When the population density is low locusts behave as individuals, much like grasshopper. As the population density is high locusts gregariously behave as band of nymphs or swarms of adults. The changes in body shape and colour and in fertility, survial and migratory behaviour are accompanied by phase change. The most important locust species is the desert locust (Schistocerca-gregaria). The international pest influences various parts of Asia, Africa and Europe. Other locust species are highly injurious to mainkind like Bombay locust and Indian migratory locust (Locusta- migratoria) found all over India in its solitary phase. The monsoon months in July and August suitable are for locust multiplication conditions. The new generation produced migrate eastward into the Indian desert areas in May and June.

The Tuppa area, which is located 10-12 km away from Nanded city, and covers an area of about 66 km² was blessed with good , quality of water before industrialisation in this area. But unplanned execution of work and excessive industrialization and many other human activities has brought us to an alarming stage. In the face to live a luxurious life style we have been destroying the natural ecological balance.

The degradation of dam has occurred not only due to influents inflow but also by sanitation, domestic sewage, agricultural waste, and other activities around the dam like washing cloths, cattle, vehicles, and dumping of solid waste. Apart from it a large number of heavy metals is contributed by corrosion of metal pipe, runoff water from surrounding area during rainy season discharge of decorative paints in water bodies.

Nanoparticle characterization is necessary to establish understanding and control of nanoparticle synthesis and applications. Characterization is done by using a variety of different techniques, mainly drawn from materials science.
